    Transforming Purification with i3 DMC Affinity Chromatography

    Affinity chromatography is one of the most selective methods for purifying antibodies and other biomolecules. With Digital Membrane Chromatography (DMC), i3 Membrane expands this proven principle to include a new form of process control: While traditional Protein A methods require a switch to acidic buffer conditions for elution, DMC makes the release of bound molecules electrically controllable.

    To achieve this, the membrane is positioned between two gold-coated electrode membranes and connected via integrated contacts to the digital control unit, the i3 DMC Control. The control unit applies a voltage to the membrane adsorbent, thereby altering local electrochemical conditions within the membrane system. These locally generated changes influence the interaction between the membrane and the target molecule, enabling its release. Elution is thus activated precisely where the binding occurs – directly on the membrane.

    This opens up new possibilities for sample preparation in early discovery and preclinical research. The technology reduces harsh process conditions, helps preserve protein integrity, and makes samples available more quickly for downstream analyses. In this way, i3 DMC addresses a key challenge in modern antibody research: Rapid purification with maximum recovery rates, without acid-induced aggregation or fragmentation.     The i3 DMC PrA 0.1 mL is a Protein A-functionalized membrane adsorber for the selective purification of monoclonal antibodies, Fc-containing molecules, and other Protein A-binding molecules. Its two-layer, open-pore cellulose membrane (0.2 µm) allows for convective flow and transports target molecules directly to the binding sites. With a bed volume of 0.1 mL and an IgG1 binding capacity of up to 5 mg, the adsorber is suitable for use in research and development applications.

    How the i3 DMC Workflow Works

    1. Integrating the Membrane Adsorber
      The selected i3 DMC membrane adsorber is integrated into the FPLC system’s fluid path via Luer-Lock connections
    2. Make the electrical connection
      The connection cables of the i3 DMC Control are connected to the integrated IN and OUT contacts of the membrane adsorber
    3. Select the protocol
      Depending on the membrane adsorber, format, and application, a single voltage, a preset, or a custom sequence is selected
    4. Trigger elution
      During the purification process, the i3 DMC Control applies the defined voltage at the appropriate time. The resulting local electrochemical reactions lead to the release of the bound target molecules
    5. Process the eluate
      The FPLC system removes the eluate from the membrane adsorber and makes it available for further processing or downstream analysis
